TREM-1 (Triggering Receptor Expressed on Myeloid Cells 1) is a critical cell surface receptor belonging to the immunoglobulin superfamily. It is predominantly expressed on blood granulocytes, monocytes, and activated macrophages, where it acts to amplify inflammatory responses triggered by bacterial and fungal infections. Beyond acute infection, TREM-1 plays a pivotal role in chronic inflammatory pathways, particularly in the pathophysiology of inflammatory bowel disease (IBD), rheumatoid arthritis, and metabolic inflammation. Interrogating the TREM-1 signaling cascade is a primary focus for therapeutic antibody screening, small molecule inhibition, and biomarker validation campaigns.
Product Features & Specifications
- Expression Host: Engineered and produced using a high-yield E. coli bacterial expression system.
- Amino Acid Sequence: Represents the extracellular domain of human TREM-1 (amino acids 21–205; UniProt ID: Q9NP99-1).
- Fusion Tags: Features a C-terminal AviTag™ peptide sequence alongside a C-terminal 6xHis tag for streamlined detection and capture.
- Precision Biotinylation: Site-specifically biotinylated at the AviTag™ sequence using BirA enzyme, preserving the native structural conformation of the binding interface.
- Molecular Mass: Resolves at approximately ~24 kDa under reducing conditions.
- High Purity: Formulated to >90% purity as verified by Coomassie blue staining on SDS-PAGE.
- Storage Buffer: Supplied in 20 mM Tris-HCl, pH 7.5, 100 mM NaCl, 10% glycerol for optimal long-term stability.
Validated Applications
This site-specifically biotinylated TREM-1 construct is optimized for robust performance in functional target-binding assays, high-throughput screening (HTS) of neutralizing biologics, surface plasmon resonance (SPR), bio-layer interferometry (BLI), and streptavidin-coated plate ELISA formats.
