Product Overview

KRAS G12V GppNHp-loaded is the active-state format of the third most clinically relevant KRAS mutation. Along with G12C GppNHp and G12D GppNHp, this product completes a three-mutant active-state panel that enables comprehensive profiling of pan-KRAS and mutation-selective inhibitor candidates across the most prevalent oncogenic KRAS variants.

This product provides recombinant human KRAS G12V (isoform KRAS4B, aa 1–185), loaded with GppNHp and site-specifically biotinylated at the N-terminal AviTag™. In the GppNHp-loaded state, KRAS G12V adopts the active switch conformation with high affinity for RAF1-RBD and other RAS effectors, consistent with the constitutively active behavior of this mutant in cells.

No other catalog supplier currently offers biotinylated GppNHp-loaded KRAS G12V as a ready-to-ship product. This format eliminates the in-lab nucleotide exchange step and provides a defined, stable active-state protein directly compatible with streptavidin-based assay formats.

Technical Notes

KRAS G12V active-state effector affinity

GppNHp-loaded KRAS G12V binds RAF1-RBD with comparable affinity to G12D GppNHp and WT GppNHp. The valine substitution does not significantly alter effector interface geometry in the active state — its primary effect is on GTP hydrolysis, not effector binding. This means that inhibitors disrupting the KRAS–RAF interface should show similar potency against G12V as against other active-state mutants, unless the compound contacts the G12 position directly.
