Technical Notes
G12V impairs GTP hydrolysis more severely than G12D
The valine at position 12 creates steric clash with the catalytic glutamine (Q61) and the arginine finger of GAP proteins, impairing intrinsic GTPase activity more severely than G12D. In cells, KRAS G12V is more constitutively GTP-loaded than G12D. In vitro, both proteins are supplied in the GDP state for practical reasons (stability), and nucleotide exchange is required to access the active state.
Pocket geometry differs from G12C and G12D
The G12V mutation creates a different switch II pocket geometry compared to G12C and G12D, which is why G12C-selective covalent inhibitors do not bind G12V and G12D-selective compounds (e.g., MRTX-1133) have reduced potency against G12V. Do not assume inhibitor cross-reactivity between mutants without experimental confirmation.
