Application: Biotinylated KRAS G12C protein was used in a TR-FRET nucleotide exchange assay to profile inhibition of SOS1-mediated nucleotide exchange by KRAS-targeting compounds.

Key assay details: The KRAS G12C assay used biotinylated KRAS protein with Tb-streptavidin, BODIPY-FL-GDP, SOS1, and GTP. Final concentrations included 10 nM KRAS G12C, 100 nM BODIPY-FL-GDP, 0.17 nM Tb-streptavidin, 50 nM SOS1, and 10 µM GTP.

Reported result: In the nucleotide exchange assay, MRTX1133 inhibited KRAS G12C with an average IC50 of 4.91 nM, and AMG510 inhibited KRAS G12C with an average IC50 of 8.88 nM.
