T4 RNA Ligase 2, also known as T4 Rnl2 (gp24.1), exhibits high activity for both intermolecular and intramolecular RNA strand joining. Unlike standard T4 RNA ligases, T4 RNA Ligase 2 is significantly more efficient at joining nicks on double-stranded RNA (dsRNA) than at ligating the ends of single-stranded RNA (ssRNA).

- RNA-to-DNA Ligation: Ligates the 3′-OH of RNA to the 5′-phosphate of DNA within a double-stranded hybrid structure (the non-ligated strand can be either RNA or DNA).

- Substrate Requirements: Requires an adjacent 5′ phosphate and 3′ OH for ligation. Requires ATP for activity unless the substrate is preadenylated on the 5′ end.

Supplied Components & Storage
Each vial is supplied with 1 mL of 10X Reaction Buffer (100 mM Tris-HCl, pH 7.5, 20 mM MgCl₂, 20 mM DTT, 5 mM ATP).
Storage Conditions: Store at -20°C in its storage buffer (10 mM Tris-HCl, pH 7.5, 100 mM NaCl, 0.1 mM DTT, 0.1 mM EDTA, 50% Glycerol).

Unit assay conditions: One unit is defined as the amount of enzyme required to ligate 50% of 0.4 µg of an equimolar mix of a single-stranded 15-mer RNA to the 5′ phosphorylated end of a 20-mer DNA in 20 µl 1X reaction buffer following a 30 minute incubation at 37°C
Storage: The enzyme and supplied components must be stored at -20°C
