Saposin C (SAP-C) is a small, heat-stable lysosomal glycoprotein derived from the prosaposin precursor protein via site-specific proteolytic cleavage. Among the four prosaposin-derived domains, Saposin C shares high structural homology with Saposin A and acts as an indispensable biological cofactor required for the lysosomal activation of β-glucocerebrosidase (GBA)—the enzyme deficient in Gaucher disease. Additionally, Saposin C plays a critical role in cellular immunology by facilitating the presentation of lipid antigens through CD1b molecules to human T cells. While native lysosomal saposins are heavily glycosylated, non-glycosylated recombinant variants expressed in bacterial systems maintain full biological activation properties in functional in vitro biochemical assays.

Beyond its role in sphingolipid catabolism, recombinant Saposin C is widely utilized in translational oncology research.
