For decades, the KRAS protein was considered "undruggable" due to its smooth, shallow surface that lacked binding sites for small molecule drugs, making it difficult to target for cancer treatment.

Molecular glues are small molecules that force interactions between KRAS proteins and other cellular proteins, often resulting in degradation or inhibition of oncogenic signaling (1, 2).

Revolution Medicines pioneered a novel class of KRAS inhibitors that utilize Cyclophilin A (CypA) to target previously "undruggable" mutant RAS cancers (1). These inhibitors act as molecular glues, binding to the abundant chaperone protein CypA. This binary complex then binds specifically to active (GTP-bound) mutant RAS, creating a new, stable "tri-complex" that inhibits tumor growth.
