Galectin-3, also known as LGALS3, is a prominent member of the beta-galactoside-binding animal lectin family. It is unique as the only chimeric galectin containing a carbohydrate recognition domain (CRD) linked to an extended N-terminal proline- and glycine-rich domain. Widely expressed by myeloid cells, macrophages, and endothelial cells, Galectin-3 is a multifunctional protein regulating cell adhesion, macrophage polarization, innate immunity, and T-cell dynamics. Its overexpression in the tumor microenvironment is strongly linked to tumor transformation, metastasis, and immune evasion, making it an essential target for cancer drug discovery and fibroinflammatory disease research.

Product Features & Specifications

  • Expression System: Engineered and expressed in an optimized E. coli bacterial expression platform.
  • Sequence Range: Recombinant human full-length Galectin-3 construct spanning amino acids 1–250 (UniProt ID: P17931).
  • Fusion Tags: Equipped with an N-terminal Polyhistidine (6xHis) tag and an AviTag™ peptide sequence for flexible detection and directed capture.
  • Precision Biotinylation: Site-specifically biotinylated at the N-terminal AviTag™ using BirA enzyme, ensuring uniform molecular orientation and protecting the binding activity of the C-terminal CRD.
  • High Purity: Evaluated at >90% purity via Coomassie blue staining on SDS-PAGE under reducing conditions.
  • Storage Buffer: Formulated in 20 mM Tris-HCl, pH 7.5, 150 mM NaCl, 1 mM 2-mercaptoethanol, and 10% glycerol for excellent structural stability.

Validated Assay Applications

This site-specifically biotinylated human Galectin-3 protein is fully optimized for robust performance in functional target-binding studies, carbohydrate-binding assays, high-throughput screening (HTS) of small molecule inhibitors, Surface Plasmon Resonance (SPR), Bio-Layer Interferometry (BLI), and streptavidin-coated plate ELISA layouts.
