Biotinylated KRAS WT in the GDP state is particularly useful for nucleotide exchange assays: SOS1 activity can be monitored by fluorescent nucleotide exchange (e.g., BODIPY-FL-GDP or mantGDP displacement) after capturing the protein on a streptavidin surface or in solution. The site-specific biotin avoids modification of exchange-relevant residues.

Technical Notes
WT KRAS hydrolyzes GTP efficiently — do not use with live GTP
Unlike oncogenic mutants, WT KRAS has intact GTPase activity (T35 and G60 positions are WT). If you add live GTP to this protein, it will hydrolyze back to GDP within minutes. For applications requiring the active conformation of WT KRAS, use the GppNHp-loaded version which is hydrolysis-resistant.
SOS1 exchange assay compatibility
GDP-loaded KRAS WT is the preferred substrate for SOS1 GEF assays. SOS1 catalyzes release of bound GDP and loading of GTP (or fluorescent GTP analogues). For a standard fluorescent exchange assay, pre-incubate KRAS WT GDP-loaded with BODIPY-FL-GDP at low concentration to label a fraction of molecules, then add SOS1 and monitor fluorescence change as labeled GDP is displaced by excess unlabeled GTP.
