Technical Notes
GppNHp vs. GTPγS vs. GTP
GppNHp (also written as GMPPNP) and GTPγS are both non-hydrolyzable GTP analogues used to lock KRAS in the active conformation. GppNHp replaces the bridging oxygen between β and γ phosphates with an imido nitrogen, preventing hydrolysis while maintaining a conformation nearly identical to GTP-bound KRAS. GTPγS substitutes a sulfur for the γ-phosphate oxygen and is slightly more reactive. For most inhibitor screening applications, GppNHp is preferred due to its greater chemical stability and the larger body of published KRAS structural data generated with this analogue. Using GTP directly is not practical for catalog supply due to hydrolysis during storage — the protein would lose activity during shipping and storage.
Why site-specific biotinylation matters for active KRAS assays
Chemical biotinylation (NHS-ester chemistry) modifies exposed lysine residues. KRAS contains multiple lysines in the hypervariable region (aa 167–185) that are important for membrane targeting, and lysines in the catalytic domain involved in nucleotide coordination (K16, K117). Random biotinylation risks modifying these residues, reducing nucleotide affinity or altering effector binding. AviTag biotinylation at the defined N-terminal tag position avoids all of these residues.
