Product Overview

KRAS G12D inhibitors under clinical development (including MRTX-1133 and RMC-9805) block the active, GTP-bound conformation of KRAS G12D. Screening these inhibitors requires constitutively active KRAS G12D in sufficient quantity and stability for high-throughput formats. This product delivers exactly that: recombinant human KRAS G12D loaded with GppNHp (guanosine 5’-[β,γ-imido]triphosphate), a non-hydrolyzable GTP analogue that locks KRAS in the active conformation without requiring live GTPase activity.

Key Features

  • KRAS G12D clinically relevant mutation
  • Supplied pre-loaded with GppNHp
  • N-terminal AviTag™, N-terminal 8XHis tagged construct
  • Site-specifically biotinylated at the N-terminus
  • High purity recombinant protein
  • Optimized for binding assays
  • Research Use Only (RUO)

Applications

  • SPR
  • BLI
  • ELISA
  • Pull-down
  • AlphaScreen
  • MST
  • Nucleotide-state-dependent inhibitor profiling
  • Effector binding assays
  • Protein–protein interaction studies
  • Drug discovery workflows

Technical Notes

GppNHp vs. GTPγS vs. GTP

GppNHp (also written as GMPPNP) and GTPγS are both non-hydrolyzable GTP analogues used to lock KRAS in the active conformation. GppNHp replaces the bridging oxygen between β and γ phosphates with an imido nitrogen, preventing hydrolysis while maintaining a conformation nearly identical to GTP-bound KRAS. GTPγS substitutes a sulfur for the γ-phosphate oxygen and is slightly more reactive. For most inhibitor screening applications, GppNHp is preferred due to its greater chemical stability and the larger body of published KRAS structural data generated with this analogue. Using GTP directly is not practical for catalog supply due to hydrolysis during storage — the protein would lose activity during shipping and storage.

Why site-specific biotinylation matters for active KRAS assays

Chemical biotinylation (NHS-ester chemistry) modifies exposed lysine residues. KRAS contains multiple lysines in the hypervariable region (aa 167–185) that are important for membrane targeting, and lysines in the catalytic domain involved in nucleotide coordination (K16, K117). Random biotinylation risks modifying these residues, reducing nucleotide affinity or altering effector binding. AviTag biotinylation at the defined N-terminal tag position avoids all of these residues.
