TL1A, also known as TNFSF15, is a tumor necrosis factor (TNF) family member cytokine predominantly expressed by monocytes, macrophages, dendritic cells (DCs), synovial fibroblasts, and endothelial cells. Its cognate cell surface receptor, DR3 (TNFRSF25), is primarily expressed on T cells, where TL1A signaling co-stimulates T-cell activation and drives pro-inflammatory cytokine production. Dysregulation of the TL1A/DR3 pathway is strongly linked to human inflammatory bowel disease (IBD), Crohn's disease, and various autoimmune pathologies, making it a high-value target for therapeutic antibody screening and biomarker validation.
