AMTAG Tn5 Transposase is a hyperactive form of Tn5 transposase engineered for rapid, robust in vitro transposition. It enables researchers to randomly insert Tn5 transposons into any target DNA, streamlining workflows by combining fragmentation and adapter addition into a single, seamless step.
For efficient transposition, the enzyme requires that each transposon contain a specific 19-bp transposase recognition sequence (Mosaic End or ME sequence: 5’–CTGTCTCTTATACACATCT–3’) at both ends. Upon binding, the enzyme forms a stable Transposome complex, catalyzing a random "cut-and-paste" reaction that yields a 9-bp sequence duplication immediately flanking the insertion site.
This unloaded formulation provides ultimate flexibility, allowing you to load any custom adapter sequence or transposon DNA of any length for tailored next-generation sequencing (NGS) applications.
Key Applications
- Custom Transposome Assembly: Flexible loading with user-defined oligonucleotide adapters.
- NGS Library Preparation: Efficient tagmentation for second-generation genomic sequencing, significantly reducing hands-on time and DNA input requirements compared to traditional methods.
