T4 RNA Ligase 1

T4 RNA Ligase 1 (ssRNA Ligase) catalyzes ATP-dependent ligation of single-stranded nucleic acids (RNA or DNA) by joining a 5' phosphoryl-terminated nucleic acid donor to a 3' hydroxyl-terminated nucleic acid acceptor through the formation of a 3'→5' phosphodiester bond.

20 mM Tris-HCl, pH 7.5, 100 mM NaCl, 1 mM DTT, 0.1 mM EDTA, 50% Glycerol

T4 RNA ligase 1 is supplied with a vial (1mL) of 10X Reaction Buffer (200 mM Tris-HCl, pH 7.5, 100 mM MgCl_2, 20 mM DTT) and a vial (0.2 mL) of 10 mM ATP.

Unit Assay Conditions                                                                                               

One unit is defined as the amount of enzyme required to convert 1 nanomole of 5´-phosphorylated 15-mer RNA into a phosphatase-resistant form in 30 minutes at 37°C in 1X T4 RNA Ligase 1 reaction buffer, supplemented with 1 mM ATP
